A pesar de esto, no se compara con el crecimiento experimentado por la Inseminación Artifi--cial (IA) durante su primera década, en los años 1940 y 1950. Actualmente la IA juega un rol importante en el desarrollo de .la --Transferencia de Embriones (TE). Aunque la Transferencia 'de embriones se ha hecho en los laboratorios y en animales de granja en forma limitada -desde 1980, la técnica fue experimental hasta la década pasada. Parece, según recientes informes, que el progreso en la congelación de los embriones y su predeterminación sexual predicha hace dos años, podría resultar en el uso comercial en gran escala de uno o de ambos procesos antes de 1985. Caracterización e identificaci6n de una virosis en lechuga (Lactuca sativa .h..:.l Agrociencia. No. 56 Pág. 163-174. 1984. RESUMEN La virosis, objeto· de este estudio, fue encontrada en las áreas aledañas a Chapingo y Xochimilco. El virus fue transmitido, mecánicamente, por el áfido Myzus persicae y por semilla. Como hospedantes del virus resultaron: Chenopo-dium amaranticolor, Gomphrena globosa, Lactuca sativa, Spinacea oleracea y Che.!!.Q.P.Odium guinea; en las dos primeras especies sólo produce lesiones locales, -mientras que en las dos siguientes produce clorosis y mosaicos bien marcados, y en la Oltima induce tanto lesiones locales como infección sistémica. El punto de inactivación tErmica del virus fue de 60ºC, el punto final de dilución----1:1000 y la inefectividad jE_ vitre se conservó por 48 horas. En preparaciones de tejido epidérmico de lechuga, previamente teñidas y vistas al microscopio de luz, se observó la presencia de inclusiones citoplasmáticas del tipo amorfo gr! nuloso, algo redondeadas y la mayoría cercana _al núcleo. La variedad que se el cogió con fines de purificación fue la Black Seed; en ella, la máxima concentr! ción del virus se encontró entre los 10-12 días posteriores a la inoculación, cuand~as plantas fueron sometidas a un fotoperfodo de 24 horas de luz con intensidad de 12 200 lux y con temperatura de 24º-25ºC. La técnica de purifica-ci6n consisti6 en licuar el tejido enfermo en una solución amortiguadora deborato 0,2 MpH 8 más ácido tioglfc6lico al 0.1% más ácido asc6rbico; filtrar a través de manta de cielo, clarificar con butanol al 8.5%, concentrar por ·ultracentrifugac4-6n y purificar totalmente con gradientes, de densidad y sacarosa al 15%. Se obtuvo un antisuero, cuyo título estimado por pruebas de precipitaci6n fue de 1:1280 y por anillo interbásico de 1:640. De acuerdo a las pruebas realizadas se concluye que el agente causal de la virosis estudiada es el virus -mosaico de la lechuga (Lettuce mosaic virus). �5 ESTUDIO COMPARATIVO DE DOS ANTIGENOS SOMATICOS DE Fasciola hepatica EN EL DIAGNOSTICO DE FASCIOLASIS EN OVINOS GOMEZ ARROYO, ALFREDO et al. Estudio comparativo de dos antígenos somáticos de Fasciola hepatica en el diagnóstico de fasciolasis en ovinos. Veteri nc¡.ria-México. Vol. XV No. 3 Pág. 193-198. 1984. RESUMEN Se prepararon dos antígenos somáticos de Fasciola hepatica adulta de -origen bovino, un extracto salino crudo (AS) y un extracto salino deslipidizado (ASO). Estos antígenos se evaluaron en las pruebas de intradennorreacción (IDR), hemoaglutinación pasiva (HP), difusión doble en agar (DD) e inmu noensayo en capa delgada (IeD), y se detenninó la sensibilidad y especificidad para cada prueba. Para esto se utilizaron 32 ovinos localizados en Tu-lancingo, Hgo., zona de alta incidencia de fasciolasis, y 50 ovinos negati-vos a F. hepatica en exámenes coproparasitoscópicos, provenientes de Hueytamalco, Pue., zona de baja incidencia. Los valores obtenidos para la sensib.1_ lidad de las distintas pruebas utilizando el antígeno AS fueron los siguientes: ÍOR, 84%; HP, 81%; reo, 56% y DO, 19%. La especificidad varió entre 70 y 96%. Con el antígeno ASO la sensibilidad de las pruebas fue: rDR, 87%; -HP, 59%; reo, 78% y DO, 6%. La especificidad varió entre 74 y 1001:. Al an~ lizar estadísticamente estos resultados se concluyó que IDR y HP son muy ad~ cuados para el diagnóstico de fasciolasis en ovinos. The producer pul. a¡ the time resulted in a 'No' vote. By 1982 thc cstimated cost of eradication had more than doubled and the second producer poli seemed teed up to produce a ·Yes' vote. The veterinary reasons for having an eradication scheme had not changed al ali, so the decision by ·. ' ., ·e Min,ístry to allow a change of course must have been a political one. However, at an estimated cost of f.6 million or so eradication still seemed a good route to follow. Rut th~t's where the scheme started to go aclrift. Not only were the costs profoundly under-estimated, various anomalies crept in. Producers voted Cor a scheme which would involve total slaughter and ¡,,yment for consequenlial ~- .. The consequential loss payments were withdrawn after the main lhrust of slaughtering and producers were advised to arrangc thcir own insura.~ce. Later a system of partía! slaughter was introduced, presum,bly. because ít is safe and savcs money, !l•11 if suc~ a ,tstem is acceptable now would it not have been acceplabl~ ear!ier· - 1.-1d saved even more money? Grac i:• a la experiencia de investigadot'es y a las inquietudes de t~cnicos especializados en reproducci6n animal que desde el a~o de 1980 (cuando Walter -Heape, en Cambridge, Inglaterra report6 el primer bito de transferencia realiz~ da en conejas) a la fecha .han trabajado en el área de la fisiologfa de la reproducd.qn; .no obstant,e ,los, j"aEasos y limitaciones anteriores, se han podido desar rollar e ir perfeccionando la técnica para llevar a cabo !l transplante de em--briones en cerdos, con dividendos satisfactorios, para que en la actualidad se realicen hasta en una forma comercial en varios países, y México no es la excepci6n, pues aunque no se tenga el transplante de embriones en fonna comercial, sí se puede aplicar la técnica para llevarlo a efecto. La prueba está en un transplante que realizaron experimentalmente en conjunto el Departamento de Reproducci6n e Inseminación Artificial y la granja experimental porcina Zapotitlán de la Facultad de Medicina Veterinaria y Zootecnia de la UNAM durante el ano 1982. cuyo logro fue positivo. Las transferencias embrionarias solamente se justifican e~ animales de un extraordinario valor genético, lo que permite obtener lechones mejorados, con la ventaja de reducir ciertas enfermedades enzoóticas por medio de la inmunidad p1siva de cerdas recept oras , adaptadas a un medio ambiente determinado. Además, la transferencia embrionaria es un proceso que nos pennite reducir notablemente el tiempo necesario para establecer una raza. Por J_Q.A:anto, se puede lugrar la �28 obtención de nuevas iíneas genéticas con su gama de ventajas debidas a la heredabilidad, en beneficio de la producción de carne de cerdo para alimento del -hombre por medio del transplante de embriones en los porcinos se podrá incremen tar la producci6n de carne de cerdo para consumo humano. 1 r'I 1 1 1 1 1 1 1 1 1 I"" r 1 1 1 1 La clave para lograr resultados positivos en el transplante de embriones consiste en la combinaci6n y ejecuci6n d~ los siguientes seis puntos básicos: -La sincronía correcta de los estros de las cerdas donadoras y las cerdas receptoras. ---Provocar la superovulaci6n en las cerdas dJnadoras. -Fertilizaci6n de los 6vulos en cerdas donadoras. --Obtención de los emb>'iones. --+lanipulación y métodos de conservación de los embrione~. -La técnica para la transferencia de embriones .~ las cerdas receptoras, la , cual puede realizarse por métodos quirúrgicos o no quirúrgicos. Uno de los puntos que resulta m~s importante es la sincronizaci6n de es--tros entre las cerdas donadoras y receptoras, pues la supervivencia de los em-briones va a depender del estado de desarrollo y las condiriones internas del útero de la hembra receptora, y los mejores resultados se han obtenido con el uso de gonadotropinas extrahipofisiarias, las cuales facilitan la detenninación de1 momento preciso del inicio de las ovulaciones. En el comercio es factible adquirir dichas hormonas, en presentación liofilizada con una concentración de 400 UI de gonadotropina cori6nica humana (HCG) y 200 UI de gonadotropina sérica de yegu~ preñado (PMSG), para ser administrada por vía intramuscular o subcuUnea. La res pues ta al tra taini an cO hormonal es va ri ab 1e, depende de 1 tipo de hormona, utilizado para provocar la ~uperovulaci6n, y de h ~usceptib111dad ind1v1 dual de cada cerda al tratamiento hormonal. Para inducir la supercvulac10n se puede administrar hormona folículoestimulante (FSH) o bién (PMSG). En México comercialmente encontramos PMSG en forna liofilizada para inyectar por vía intramuscular, en concentraciones de 1000 U! por dosis. Cabe mencionar que con d_Q sís menores a óOO U! de PMSG los resultados de superóvulación en cerdas no han sido alentadores. 1 1 1 1 Una gran ventaja o diferencia entre administrar FSH y PMSG, es que la PMSG se administra durante el principio de la fase folicular (entre el 15° y 16° día del ciclo estral) en dosis Onica; mientras que para administrar FSH es necesa-rio intervenir varios días a la cerda, lo cua ·1 aumenta el empleo de la mano de obra y por ende el estrés en la cerda. 1 1 1 ·¡ 1 1 1 1 1 1 1 1 1 La cécnica más apropiada para el transplante de embriones en cerdas es la misma rlescrita por Hancock y Hovell en 1962, y por Dziuk, Polge y Rowsor en 1964. r.l transplante de embriones se debe realizar en un momento apropiado para la rf'cJp¡,raci6n de los embriones, el cual es entre el segunoo y qu;rto día posterior a la fertilización. Se pueden recuperar los embriones 48 horas des-pués de la monta o inseminación artificial si se hace el lavado a nivel del ovi dueto, dirigiendo el medio en sentido de los oviductos hacia los cuernos uterinos, con un exito de recuperación del 64 por ciento aproximadamente. Los embriones de la cerda donadora se recuperan por medio de métodos qui-rúrgicos, es decir, se pone bajo anestecia total a la donadora, en la que es~ cesario realizar una laparotomía para exponer los órganos genitales. Ambos --cuernos uterinos son lavados con PBS o TCM-199 a 37ºC. Aproximadamente a 10 cm de la unión útero-tubárica se realiza una pequeña incisi6n para insertar un catéter de Folley que se insulfa para no permitir el reflujo del medio hacia el exterior; a través del catéter se inyecta el medio (40 a 60 ml) inundando los óvulos fertilizados, dando un ligero masaje de arriba hacia adelante; posterio_!: mente se procede a recuperar el medio con los embriones por medio del mismo catéter el cual es recibido en vidrios de reloj estériles . . ' ' Los primeros intentos para conservar embriones durante largos períodos -fueron reportados por Chang en el año de 1971. Pero realmente Audrey y Smith presentaron su primer conferencia sobre el tema, en el año de 1952; mencionando la sobrevivencia al congelamiento y descongelarrriento de los embriones de ra t6n y rata. Polge relata en 1977 la corta historia del congelamiento de em--briones y de ese entonces a la fecha son bien conocidos los avances no salame_!! te del congelamiento del semen, sino también de la micromanipUlaci6n embrionaria, lo cual puede permitir el sexado de los embriones y quizás en un futuro no muy 1eja·no su el onifi caciOn.
